Engineered Human Ribonuclease Inhibitor With Enhanced Oxidation Resistance
Human ribonuclease inhibitor (hRI) protein inhibits the activities of angiogenin (ANG), whose activities are present in cancer cell development. hRI can be easily oxidized and going through conformational change to degrade itself due to its biochemical composition on molecular level. As the relationship between its oxidation resistance and its ANG binding and inhibitory activities remains unclear, I propose new designs of hRI variants plasmids that encode for hRI with enhanced oxidation resistance via free energy estimation and future wild-type and mutant hRI protein expression and purification scheme.
